The main character, Meg, is female as are the three alien beings who assist her in her quest (admittedly the quest is to find her father). As such, four of the six main characters are female.
There are numerous discussions between Meg, Mrs Whatsit, Mrs Which and Mrs Who. The majority of these advance the plot. Because the main plot device is "rescue your father" many of the discussions involve men, but plenty do not, also.
For specificity, during the scene where the three aliens take their leave of Meg, Calvin and Charles Wallace, Mrs Whatsit tells Meg that "I give you the gift of your faults". This dialogue is Mrs Whatsit talking to Meg about Meg herself and helping Meg to accept who she is.
There are four named female characters in this scene who spend a good deal of time talking to each other. This movie would pass the test in entirety many times over.
There are two other named female characters, Meg's mother Mrs Murray and the next-door-neighbour Veronica. Mrs Murray also talks to Meg, sometimes about her father and brother and sometimes about Meg herself. Veronika says little, and mostly is talking about Meg's father.
